Separation and purification of flavonoids from Prunella Vulgaris Linn by macroporous adsorption resin

Abstract

The characteristics of macroporous adsorption resins D-101,AB-8,X-5,DA-201,HZ-806,and HZ-801 on adsorption and desorption of total flavonoids from Prunella Vulgaris Linn were investigated.With the yield of flavonoids as index,the optimum process of separating and purifying flavonoids was screened by static and dynamic adsorption tests.The results of static adsorption test exhibited that AB-8 resin has higher absorption and desorption capacity.The optimum technology parameters of adsorption and desorption are pH 3.0~3.5,sample volume 3BV,sample concentration 1.8mg/mL,flow velocity 6BV/h.total flavonoids adsorbed on the column can be eluted by 70% alcohol at 6BV/h flow rate and at the volume of 6BV.Compared to the 41.58% of total flavonoids in crude extracts,the recovery and purity of total flavonoids were 92.36% and 89.09% respectively by the fresh resin.The recovery and purity of total flavonoids were over 80% and 60% respectively after resin used for four times.
