Abstract
[Objective] The purpose was to analyze the effects of polyphenol substance content in Castanopsis fissa on explant browning in initial culture.[Method] The browning caused death of C.fissa explant was observed by inoculating C.fissa explant on the MS medium with different concentration.of the active carbon and ascorbic acid.Meanwhile,the polyphenol substance contents in the samples of C.fissa with different ages in various positions were determined,its effects on explant browning of initial culture were analyzed and the measures of reducing explant browning in initial culture for C.fissa were researched preliminarily.[Result] The polyphenol substance contents in the samples of C.fissa in different organs and different positions were varied,and the polyphenol substance contents had extremely significant positive correlation with explant browning in initial culture of C.fissa(r=0.899).Adding 1.0-3.0 g/L active carbon and 0.1-0.3 mg/L ascorbic acid in medium was in favor of decreasing the browning of explant of C.fissa in initial culture,and the effect of active carbon was slightly better than that of ascorbic acid.[Conclusion] The browning of explant of C.fissa in initial culture was closely related with polyphenol substance contents.
