Abstract

This paper used Paeonia ostii var liishizhenii as experiment material.Compared the effect of different explants,basal mediums and combinations of hormones to callus inducing;the results indicated,to use cotyledon and radicel as explant,on the medium: 1/2 modified WPM+2,4-D 1.0 mg·L-1+ 6-BA1.0 mg·L-1+Casein hydrolysate 500 mg·L-1+L-glutamine 150 mg·L-1+sucrose 30g·L-1+ager 7 g·L-1,inducing rate reached to100%,In the research of the effect of basal medium,different concentration of inhibitor and culture condition to the browning,the result implied,browning was slighter in the basal medium 1/2 modified WPM;antioxidant and sorbent had remarkable browning prohibited effect,PVP and Phytagel took on the best browning prohibited effect in all of them.The browning index was 31.11% and 30.00%.Different culture conditions affected browning greatly,darkness,low-temperature and short subculture cycle can reduce browning in different degree.
