Abstract
Objective: To observe the changes of cyclin kinase inhibitor p27 expressed by glomerular mesangial cells in response to IL-13? ET-1? methylprednisolone and heparin sodium. Methods: IL-13(10 ng/ml and 100 ng/ml), ET-1(10-7 mol/L), methylprednisolone (0.5 μg/ml) and heparin sodium (100 mg/L, 200 mg/L) acted on cultured glomerular mesangial cells and p27 level was assessed by flow cytometry. Results: The levels of p27 in IL-13 groups were (46.74±3.25) 10 ng/ml and (23.8±2.56) 100 ng/ml, which were increased significantly compared with that of controls (P 0.001, P 0.05). Stimulating mesangial cells with ET-1 for 14 and 18 hours, the levels of P27 were lower than that of controls (P 0.05, P 0.05). Expressions of P27 with methylprednisolone for 48 and 72 hours were (9.90±9.12) and (9.12±0.93), was significantly lower than that of controls (P 0.01). Expressions of p27 in heparin sodium group (200 mg/L, 100 μg/L) were increased significantly compared with that of controls (P 0.05). Conclusions: Cyclin kinase inhibitor p27 is a very important factor in regulating glomerular mesangial cell proliferation.
