Experimental Study of an Aplastic Anemia Model Induced by Benzene and Cyclophosphamide in Mice

Abstract

Objective To establish a model of aplasfic anemia induced by benzene and cyclophosphamide in mice. Methods Kunming male mice were injected with benzene+corn oil(2.0ml/kg) once every two days and with cyclophosphamide(50mg/kg) once a day for a week.The parameters tested were as follows:general condition,peripheral blood smear,bone marrow smear,bone marrow nucleated cell count,and histopathology of liver,spleen and tibia. Results Compared with control group,experimental group mice showed the following results: leukocyte(42.4%,P0.01),erythrocyte(18.4%,P0.05),platelet(15.4%,P0.05),reticu-locyte(23.5%,P0.05) and bone marrow nucleated cell count 24.4%(P0.05) were decreased remarkably.The bone marrow smears and the patho-morphological examinations showed that hematopoietic cells were decreased with the increase of the non-hematopoietic cells,such as fat cells. These parameters indicated that the model was set up successfully. Conclusions An experimental aplastic anemia model in mice is established simply and easily with a high succsess rate and the results are stable.As a result,an experimental aplastic anemia model induced by benzene and Cyclophosphamide in mice is preliminarily set up.
