CD8~+/CD28~+ lymphocyte and their subtypes in the peripheral blood of patients with chronic hepatitis B

Objective To explore the relationship between the the CD8+/CD28+ lymphocyte and their subtypes in the peripheral blood and the clinical condition or the HBV replication of chronic hepatitis B.Methods The CD45R0、CD45RA and CD28 of peripheral blood CD8+ lymphocyte were detected by flow cytometry with multicolor fluorescence technology in 280 chronic hepatitis B cases, 28 HBV asymptomatic carriers (ASC) and 22 healthy volunteers.Results (1)The percentage of CD8+CD28+ lymphocytes of ASC (10.64±5.09%) was significantly lower than that of chronic hepatitis B and controls. There was no significant difference in CD45RO and CD45RA expression at the CD8+ lymphocytes of all groups.(2)The CD8+/CD45RA+/CD28+ in chronic hepatitis B (10.99±7.33%) group was significantly higher than that of control, while the CD8+/CD45RA+/CD28+ in chronic hepatitis B and ASC group were lower than that of control. (3)The CD8+/CD45RO+/CD28+ in HBeAg negative group was higher than that of HBeAg positive group.But there was no significant difference in CD8+/CD45RA+/CD28+.Conclusion The lower expression of the costimulation CD28 in the CD8+ lymphocytes are relative to the HBV asymptomatic carriers. The elevation of CD8+/CD28+CD45RO+ subtype was probably related to the progress of chronic hepatitis B and the serum HBeAg conversion.
