Abstract
A modified CTAB method of genomic DNA extraction of Aquilaria sinensis(Lour.)Gilg was established based on the classic CTAB method. The comparison of modified CTAB method and Kit method showed that the quality of the genomic DNA extracted by the modified CTAB method was better than that of Kit method. The OD-(260)/OD-(280) ratio of the genomic DNA extracted with modified CTAB method was between 1.8 and 2.0 and the DNA yield was about 172 μg/100 mg dry leaf. The DNA extracted with the modified CTAB method could be digested completely by restriction endonuclease to be used for downstream studies. Conclusion: The modified CTAB method was suitable for the extraction of the genomic DNA of Aquilaria sinensis(Lour.)Gilg..
