Genomic DNA Extraction and Analysis of Mesona chinensis Benth.

Abstract

[Objective] The research aimed to provide genomic DNA of Mesona chinensis Benth.for molecular studying.[Method] Genomic DNA extracted with modified CTAB method was tested for its concentration and quality,through ratio of OD260/OD280 and agarose gel electrophoresis(AGE)and RAPD-PCR,in order to study the extraction effect.[Result] Ratio of OD260/OD280 of genomic DNA,showed by means of modified CTAB,was among 1.6-2.0.The DNA strip was clear,and the integrity was well with higher purity.The molecular weight was similar with that of λDNA.The stable amplified strips could be obtained through RAPD-PCR amplification with template of the extracted genomic DNA.[Conclusion]The modified CTAB method was favorable for extraction of genomic DNA of Mesona chinensis Benth.
