Effect of recombinant human erythropoietin on tumor necrosis factor-α gene expression after spinal cord injury in rats

Abstract

Objective:To study the effect of recombinant human erythropoietin (rHuEPO)on tumor necrosis factor-α (TNF-α ) gene expression after spinal cord injury(SCI) in rats .Method:One hundred and two SD rats were divided randomly into four groups:sham operation group,spinal cord injury group,group of spinal cord injury treated with NS and group of spinal cord injury treated with rHuEPO.The modified Allen’s model of spinal cord injury was established.The expression of TNF-α mRNA after SCI was measured by reverse transcription polymerase chain reaction (RT-PCR).Results:The expression of TNF-α mRNA was detected in non-injured spinal cord,significantly increased and peaked at lh after SCI,remained significantly higher than that in the non-injured spinal cord for 24h post-injury.rHuEPO significantly reduced the expression of TNF-α mRNA in the injured spinal cord in comparison to that of control animals receiving NS vehicle at 6,12,24h after SCI.Conclusion:rHuEPO significantly reduces the expression of TNF-α mRNA after SCI in rat.
