Abstract

Objective:To investigate the effect of puerarin on the expression of Caspase-3 and Bcl-2 and the apoptosis of hepatocytes in rat's hepatic ischemia reperfusion injury.Methods:A rat model of partial hepatic ischemia-reperfusion was found.The Caspase-3 and Bcl-2 expression were measured with immunohisto-chemical method.The apoptosis of hepatocytes were observed under electron microscope.Results:Universal hepatic cell apoptosis was found after reperfusion.The expression of Caspase-3 increased significantly,and the expression of Bcl-2 was lower at 3h after reperfusion,returned to normal level at 6h and continued to increase until 48h.When treated with puerarin,there was much less apoptosis,the expression of Caspase-3 decreased obviously and the positive Bcl-2 was significantly higher.Conclusions:Puerarin can protect the liver from the ischemia reperfusion injury by increasing Bcl-2 expression and inhibiting Caspase-3 expression,alleviating the apoptosis of hepatocytes.
