Effects of PPAR-γ activation on rat mesangial cell extracellular matrix accumulation induced by advanced glycation end products

Abstract

Objective To investigate the role of peroxisome proliferator-activated receptor-γ(PPAR-γ) activation in rat mesangial cell extracellular matrix(ECM)accumulation induced by advanced glycation end products(AGEs).Methods Rat mesangial cells were treated with AGE-modified bovine serum albumin(AGEs) or native bovine(serum) albumin(BSA).The effects of AGEs(0,12.5,25,50,100 and 200 mg·L~(-1)) on mesangial cells fibronectin and collagen Ⅳ protein contents and the role of PPAR-γ activation in rat mesangial cells ECM accumulation induced by AGEs(100 mg·L~(-1)) were investigated.Fibronectin,collagen Ⅳ protein contents were detected by ELISA and PPAR-γ mRNA was analyzed by reverse transcriptase-polymerase chain reaction(RT-PCR).(Results Mesangial) cells expressed PPAR-γ mRNA.Compared with respective BSA groups,AGEs((12.5-200 mg·L~(-1)))increased fibronectin and collagen Ⅳ protein contents of rat mesangial cells.PPAR-γ activation(1.25-10 mmol·L~(-1)) could ameliorate collagen Ⅳ protein increasing induced by AGEs(100 mg·L~(-1)) in mesangial cell medium.Conclusion PPAR-γ activation can ameliorate mesangial cells collagen Ⅳ increasing induced by AGEs.
