Abstract

Objective To investigate the effects of advanced glycation end products (AGEs) on matrix metalloproteinase-2 (MMP-2) activity and expression in rat mesangial cells. Methods Rat mesangial cells were treated with 100 mg·L -1 AGE-modified bovine serum albumin(AGEs group)or native bovine serum albumin(BSA group)for 48 h. Normal mesangial cells without any treatment were as a control (DMEM group). MMP-2 activity and mRNA were analyzed by zymography analysis and reverse transcriptase polymerase chain reaction (RT-PCR), respectively. Results Both MMP-2 activity and mRNA expression of mesangial cells in AGEs group were reduced significantly, compared with BSA or DMEM group (P0.01). Conclusion AGEs can reduce the activity and expression of MMP-2 in rat mesangial cells.
