Abstract
Objective: To develop a method for determination of trace selenium in rat tissues by microwave digestion-graphite furnace atomic absorption spectrometry(GFAAS).Methods: Microwave Digestion Treatment of samples with nitric acid as the digestion agent for microwave digestion;palladium chloride is used as a matrix modifier for determination of trace selenium. Results: The best ashing temperature was 900 ℃,the atomization temperature was 2500 ℃;brain and lung tissue relative standard deviations(RSDs) were 2.83% and 3.05%,respectively;sample recoveries were in the range of 97% to 105%.Conclusion: The method for the detection of rat tissue selenium content is simple, fast, with good accuracy, and these results are satisfactory. And thus, the method can be applied to many organs and tissues of organisms in determination of the content of selenium and analysis.
