Abstract
The stem segments with axillary buds of BA-29 (Cydonia oblonga Mill.) were used as experimental explants to study the different disinfection methods and the effects of hormone on adventitious bud occurrence and multiplication.The results indicated that:(1)During primary culture process,the medium of 1/2MS+6-BA 1 mg·L-1+IBA 0.1 mg·L-1+GA 0.2 mg·L-1 was the best for induction and multiplication ,and the explants sterilized in 0.1% HgCl2 for 5min got the best result with the lower contaminate rate of below 20% and the highest survival rate of above 70%.(2)The optimal multiplication medium in subculture was MS+6-BA 1 mg·L-1+IBA 0.4 mg·L-1+GA 0.05 mg·L-1,the multiplication rate was above 3.8,and the new bud has strong growth potential.(3)1/2MS+NAA 0.5 mg·L-1+IBA 0.5 mg·L-1 was the best medium for rooting,with a rooting rate of 81% and 7.6 roots on average.After acclimatization for one week,the survival rate was up to 80% on 30 days after transplanted into sterilized medium.
