Abstract
Objective: To investigate the inhibitory effect of protocatechuic acid and other compounds on HBsAg,HBeAg,HBV DNA and its toxicity to cultured hepatocarcinoma cell line HepG2.2215 transfected with hepatitis B virus DNA. Methods: Twenty four hours after HepG2.2215 cells were vaccinated, different concentrations of drugs were added into culture fluid respectively. After the cells were treated for 4 d, culture fluid was collected and the contents of HBsAg and HBeAg were determined by ELISA. HBV DNA was analyzed by Southern blot. Results: ELISA showed that 4 compounds of 6 inhibited the expression of HBsAg and HBeAg. Inhibitory effect of 70 μg/ml protocatechuic acid on the secretion of HBeAg was stronger than that on HBsAg. Southern blot showed that 70 μg/ml protocatechuic acid could inhibit the expression of HBV DNA up to 46.54%. Conclusion: Protocatechuic acid has strong inhibitory effect on HBV in vitro , and this effect may be connected with the inhibition of HBV DNA. [
