Abstract

Objective To investigate the effect of moxibustion on type Ⅰ,Ⅲ and Ⅳ collagen synthesis in conlonic fibroblasts(CFB) and explore the mechanisms of its preventive and therapeutic actions on colonic fibrosis in ulcerative colitis fibrosis rats. Methods A rat model of ulcerative colitis fibrosis was made by an immunological method plus local stimulation. Rats were randomly divided into normal,model,herb-partition moxibustion, mild moxibustion and salicylazosulfapyridine(SASP) gavage groups. The herb-partitioned moxibustion and mild moxibustion groups were treated by herb-partitioned moxibustion and mild moxibustion, respectively,on points Qihai(CV 6) and Tianshu(ST 25,bilateral) . The SASP gavage group was treated by salicylazosulfapyridine gavage. Colonic fibroblasts were isolated and cultured in all the groups of rats. The effect of moxibustion on type Ⅰ,Ⅲ and Ⅳ collagen synthesis in colonic fibroblasts was investigated by western-blotting in the rats with ulcerative colitis fibrosis. Results Type Ⅰ,Ⅲ and Ⅳ collagen synthesis in colonic fibroblasts increased significantly in the rats with ulcerative colitis fibrosis. Type Ⅰ and Ⅳ collagen synthesis in colonic fibroblasts was significantly inhibited in both Herb-partitioned moxibustion and mild moxibustion groups of rats. Type Ⅰ collagen synthesis in colonic fibroblasts was significantly inhibited in the SASP gavage group of rats. Conclusions Moxibustion can regulate CFB collagen synthesis in ulcerative colitis fibrosis rats.
