Abstract
[Objective] To observe the distribution of glial-cell line derived neurotrophic factor (GDNF) protein and mRNA in rats brain with cerebral hemorrhage and effect of Naoyi-an on them. [Methods] The intracerebral hemorrhage(ICH) animal models in rat were established by microinjection 0.4 u Ⅶ collagenase into the globus pallidus, We observed the expresion variation of GDNF protein and mRNA at 2 h, 6 h, 1 d, 4 d, 7 d and 14 d after cerebral hemorrhage by immunohistochemistry and in situ hybridization technique, and positive cell counting served as the observation index. [Results] GDNF protein mainly expressed in microglia around the perihematom after ICH, began to express at 2 h after cerebral hemorrhage, and rose at 6 h, and peaked at 1 d. dicreased to nomal level at the 7th day and disappeared at 14 th d after ICH injury. At 1 d and 4 d, positive cell counting of the two groups had significant difference (P 0.05), GDNF mRNA were mainly expressed in neurons, reached the peak after 1 d, then decreased after 4 d of ICH injury,and decreased continuely but yet higher than the normal level after 7 d, and come around to the normal level in cells quantity and expression level after 14 d ICH injury. At 6 h, 1 d and 4 d, positive cell counting of the two groups had significant difference(P 0.01). [Conclusion] Naoyi-an can promote the expression of GDNF protein and mRNA after ICH.
