Abstract
Objective To observe the distribution of glial-cell line neurotrophic factor(GDNF)protein and mRNA in rats brain with cerebral hemorrhage.Methods The intracerebral hemorrhage(ICH)animal models were established by microinjection of 0.4u Ⅶ collagenase into the globus pallidus and observed the expression variation of GDNF protein and mRNA at 2h,6h,1d,4d,7d and 14d after cerebral hemorrhage by immune histochemistry and in situ hybridization technique,and positive cell counting served as the observation index.Results GDNF protein mainly expressioned in microglia around the hematom after ICH,began to express at 2h after cerebral hemorrhage,and peaked at 1d,and then decreased to the basal level on 14d after ICH injury.At 6h and on 1d,4d and 7d,positive cell counts of the three groups had significant difference(P0.01).GDNF mRNA was mainly expressed in neurons of cerebral cortex,hippocampus,corpus striatum,basal forebrain,substantia nigra,corpus callosum and around the hematom.Then reached the peak after 1d,and decreased after 4d of ICH injury,and to come around the normal level in cells quantity and expression level 14d after ICH injury.At 6h and on 1d,4d and 7d,positive cell counts of the three groups had significant difference(P0.01).Conclusion The expression of GDNF protein and mRNA increased after ICH.
