Cytotoxicity effect and Bcl-2 expression of K562 cells treated with hyperthermia and adriamycin in vitro

Abstract

[Objective] To investigate the inhibitive effect,the apoptosis and Bcl-2 expression of K562 cells with hyperthermia and adriamycin.[Methods] The working concentration of adriamycin against K562 was determined by MTT assay.Then hyperthermia and chemotherapy were used singly or concurrently and the cell survival rates were obtained at 48h.The inhibitive effect was evaluated by MTT.The apoptosis rates and Bcl-2 expression of K562 were determined by flow cytometric analyses.[Results] The concentration adriamycin in the experiment was defined as its IC50.The hyperthermia of 40℃,42℃ for 60min showed obvious inhibition to K562 cells at 48h(P 0.01).Adriamycin chemotherapy combined with hyperthermia showed obvious inhibitive effect o K562(P 0.01).According to flow cytometric analyses,the treatment of hyperthermia and adriamycin used alone or concurrently could obviously increase the apoptosis rates(P 0.01)and down-regulate Bcl-2 expression of K562(P 0.01).[Conclusions] Adriamycin chemotherapy combined with 60min of hyperthermia showed obvious inhibitive effect to K562 at 48h.The effect can increase the apoptosis rates and down-regulate Bcl-2 expression.
