Abstract
Objective To investigate the suppression effect of adriamycin combined with hyperthermia on chronic leukemic K562 cell line.Methods The working concentration of adriamycin against K562 was determined by MTT assay.Then hyperthermia and chemotherapy were used singly or concurrently and the cell survival rates were calculated at 48h.The suppression effect was evaluated by MTT assay.The apoptosis rates of K562 were determined by flow cytometric analyses.Results The concentration of adriamycin in the experiment was defined as its IC50.The 40℃and 42℃ hyperthermia used for 60 min showed obvious inhibition to K562(P0.01),adriamycin chemotherapy or hyperthermia both obviously inhibited the growth of K562(P0.01).Adriamycin chemotherapy combined with hyperthermia showed obvious effect to K562.According to flow cytometric analyses,hyperthermia and adriamycin used alone or concurrently all could obviously increase the apoptosis rates of K562(P0.01).Conclusion Adriamycin chemotherapy combined with hyperthermia used for 60min canobviously increase suppression effect to K562.The synergism might increase apoptosis rates of K562.
