Genotypic subtypes detection of human papillomavirus by flow-through hybridization in cervical diseases and its significance

Abstract
Objective:To evaluate the clinical application value of flow-through hybridization in identifying of human papillomavirus(HPV) genotypes for etiological diagnosis of cervical diseases.Methods:236 cases of local women were subdivided into four groups according to their histological diagnosis(31 cases of cervical cancer,10 cases of cervical intraepithelial neoplasia(CIN),118 cases of chronic cervicitis,and 77 cases of normal histological manifestation).Cervical cells prepared from 236 cases were used to detect the HPV genotypes by flow-through hybridization and the composition and distribution pattern of HPV genotypes within four group were analyzed.Results:Among 236 cases of samples,72 cases were detected to have HPV infection,from which the positive cases in cervical cancer,CIN,chronic cervicitis,and normal groups were 29(93.5%),5(50.0%),30(25.4%)and 8(10.4%),respectively.The detection rate of HPV infection among four groups were significantly different(χ2=76.041,P0.001).The percent distribution of detecting HPV genotypes were HPV-16(33.3%),HPV-52(16.1%),HPV-58(8.0%),HPV-31(8.0%),HPV-18(6.9%),HPV-11(6.9%),CP8304(3.4%),HPV-33(3.4%),HPV-53(2.3%),HPV-59(2.3%),HPV-68(2.3%),HPV-66(2.3%),HPV-6(1.1%),HPV-39(1.1%),HPV-56(1.1%),HPV-42(1.1%),HPV-43(1.1%),HPV-45(1.1%).The ratio of high-risk HPV genotypes was increased with aggravation of cervical lesion(χ2=61.190,P0.001).The 3 most common genotypes of HPV in the patients' groups were all high-risk HPV genotypes.Conclusion:Cervical diseases have close relationship to HPV infection and cervical carcinomas are mainly resulted from high-risk HPV genotypes' infection.Flow-through hybridization can become an ideal approach to determining the HPV genotypes and should be promoted for use during etiological diagnosis of cervical diseases.
