Effect of dexamethasone on proliferation and apoptosis in glioma C6 cells

Abstract

Objective To investigate the effect of dexamethasone(Dex) on proliferation and apoptosis in glioma C6 cells.Methods The glioma C6 cells were divided into 4 groups.The cell number in each group was 1.25×106.Group B、C、D were cultured with Dex,except group A.The final concentraion of Dex in B、C、D group was 10-5,10-4,10-3mol/L.The number of C6 cells were counted by ordinary counting chamber,the cell cycle and the apoptotic rate of C6 cells were detected by flow cytometry.Results After cocultured for 24 h,the number of C6 glioma cells in group A,B,C,D were 4.37×106,4.29×106,3.57×106 and 3.44×106,there were remarkable differences between group B,C,D and group A(all P0.05).The apoptosis rate in these 4 groups were 0.37%,0.52%,0.39% and 8.24%,there were remarkable differences between group A,B,C,D and group D(all P0.05).The proportion of C6 cells in the phase G0/G1 were 82.42%,93.21%,93.71%,77.52%,in the phase S were 13.22%,5.38%,4.06%,14.74%,in the phase G2/M were 4.36%,1.41%,2.23%,7.74%.There were remarkable differences between group B,C and group A in the phase G0/G1,S(all P0.05).There was remarkable difference between A,B,C group and D group in the phase G2/M(all P0.05).Conclusion Dex can inhibite cell proliferation and induce apoptosis by inhibiting C6 cell cycle progression.
