Abstract
Objective To observe the different temperature of thermo-chemotherapy to the BIU-87 cells of bladder cancer cell line, including proliferation and apoptosis, and to explore its possible mechanism. Methods The logarithmic growth phase BIU-87 cells were divided into four groups, group A was the control group, group B was thermo-chemotherapy group one[(38±0.2)℃], group C was thermo-chemotherapy group two[(40±0.2)℃], group D was the thermo-chemotherapy group three[(43±0.2)℃], with four methyl thiazolyl tetrazolium(MTT) assay, BrdU incorporation experiment to detected the effects on cell growth and the proliferation roles. Flow cytometry(FCM) were measured on the influence of cell cycle and apoptosis. Western blot(Western blot) were to detected the expression of the protein of BcL-2 and Capase-3. Results MTT assay showed relative to the control group, group B, C, D can led to inhibition of growth and proliferation, the inhibitory effect of group D was the most obvious, the difference between the test group and the control group was significant(P0.05). BrdU incorporation experiments suggested that the proportion of cells in S phase in group B, group C, group D was significantly less than in group A(P0.05), where D is the lowest percentage of the group. FCM showed that the BIU-87 after the treatment of 48h in experimental group had obvious changes in cell cycle, the percentage of G0/G1 phase cells increased, the percentage of S phase cells decreased, especially in group D the most obvious change, compared with the control group, the difference was statistically significant(P0.05). Compared with the control group, the apoptosis rate was significantly higher in experimental group, the D group was the most obviously Western blotting results showed that in the experimental group, the expression of Caspase-3 protein was increased, and the decreased expression of BcL-2 protein. The most obvious change was in group D. Conclusion The thermo-chemotherapy of GEM can inhibit the proliferation of BIU-87 cells, by down-regulating the expression of BcL-2 protein and increased the expression of Caspase-3 protein activity, thereby inducing cell apoptosis. And as the temperature increased, the inhibition was more significantly.
