Determination of triptolide and two alkaloids in the extract of Tripterygium wilfordii by HPLC

Abstract

Objective To develop a high performance liquid chromatography (HPLC) method for the determination of triptolide,wilforgine and wilfornine F in the extract of Tripterygium wilfordii Hook. f. Methods The HPLC separation was carried out on a YMC-Pack ODS-A column (4.6 mm×150 mm,5 μm) with acetonitrile-0.02 mol/L sodium dihydrogen phosphate as the mobile phase for gradient elution. The flow rate was 1.0 mL/min,the detection wavelength was 210 nm,and the temperature of column was 25 ℃. Results Linearity was confirmed for triptolide,wilforgine and wilfornine F. The average recoveries were in the range of 98.81%-99.84% with RSDs less than 1.42% (n=6). Conclusions The method is accurate,repeatable and applicable to the determination of triptolide,wilforgine and wilfornine F in T. wilfordii.
