Determination of ginkgolides in ginkgo leaf extract and dipyridamole injection by HPLC-ELSD

Abstract

Objective To establish an HPLC-ELSD method for the determination of biloblide,ginkgolide A,ginkgolide B,and ginkgolide C in the ginkgo leaf extract and dipyridamole injection.Methods The chromatographic conditions included Hypersil ODS2 column(200 mm×4.6 mm,5 μm),THF-methanol-water(1∶22∶77) at the flow rate of 1.0 mL·min-1 of the mobile phase and at(35±0.1) ℃.ELSD was used with the drift tube temperature of 94 ℃ and the gas pressure of 50 psi.Results The calibration curve was linear over 4.25~16.5 μg for biloblide,3.74~14.52 μg for ginkgolide A,3.58~13.86 μg for ginkgolide B,and 3.40~13.20 μg for ginkgolide C.The average recoveries were 100.3%(RSD=0.76%),100.0%(RSD=0.39%),99.8%(RSD=0.48%) and 99.9%(RSD=0.70%),respectively.Conclusion The HPLC-ELSD method was precise,reproducible and stable,which can be applied in the quality control of the ginkgo leaf extract and dipyridamole injection.
