Modification of microspore in vitro culture of Brassica napus L.

Abstract

Brassica napus L. microspores from F1 derived from low oil content line 1064 and high oil content line H105 hybrid were cultured for establishing double haploid (DH) population. F1 were planted in field in Nanjing and Xining,respectively. Embryogenesis rate difference was observed between two locations. Regeneration rate was recorded using embryos with sizes of 0.4cm to 0.6cm,0.6cm to 0.8cm and 0.8cm. Doubling efficiency of three colchicine treatment methods were also investigated. Results showed that embryogenesis rate was higher and reached up to 8.6 embryos per plate in Xining than that in Nanjing. 0.6cm to 0.8cm sized embryos transferred to B5 solid medium gave a higher regeneration ratio of 48.53%.High doubling efficiency of 46.23% was obtained from 50 mg/L colchicine suspension treatment of microspores for 48h.A DH population of 123 lines was obtained in this study. It can be used as the basic population for QTL analysis of oil content in B.napus L.
