Abstract

[Objective] To observe the proliferation inhibitive effect of grub extract on human breast cancer MCF-7 cell line.[Methods] MTT assay was used to measure the growth inhibition rate of human breast cancer MCF-7 cell line treated with the grub extract. The expression of PCNA,Ki-67 and CyclinD1 was detected by immunohistochemistry before and after exposure to the grub extract.FCM was used to assay the change of cell cycle.[Results] The growth inhibition rate of MCF-7 cells treated with the grub extract was significantly higher than that of control, and the inhibitive effect was time and concentration dependent. The expression of Ki-67,PCNA and CyclinD1 was down-regulated and the cells were cumulated in S phase.[Conclusion] The proliferation of human breast cancer MCF-7 cell line can be inhibited significantly by the grub extract in vitro. It is related with down-regulation of CyclinD1,Ki-67 and PCNA.
