Effect of Nodosin on Nitric Oxide and Cytokines Release from RAW264.7 Cells Induced by Lipopolysaccharide

Abstract

Objective To observe the effect of nodosin on nitric oxide(NO) and cytokines release from RAW264.7 cells induced by lipopolysaccharide(LPS).Methods Methyl thiazolyl tetrazolium(MTT) assay was used to test the influence of nodosin on the proliferation of in-vitro cultured murine macrophage cell line RAW264.7.The secretion of cytokines from RAW264.7 cells was stimulated by LPS,and then was intervened with nodosin at the concentrations of 1.25,2.5 and 5.0 mmol/L.After intervention,the level of NO in the culture supernatant was tested by Griess Reagent,and the levels of interleukin 6(IL-6),IL-10,tumor necrosis factor alpha(TNF-α) were determined by enzyme-linked immunosorbent assay(ELISA).Results Nodosin had no effect on proliferation of RAW264.7 cells within 10 mmol/L,but showed dose-dependent effect on inhibiting the release of NO,IL-6 and TNF-α,and on increasing the production of IL-10 in the LPS-stimulated RAW264.7 cells.Conclusion The anti-inflammatory action of nodosin is probably related with the inhibition of inflammatory factors of NO,IL-6 and TNF-α,and with the increase of IL-10 expression.
