Abstract
【Objective】To investigate the influence of triptolide A(TA)on the production of inflammation-promoting and anti-inflammatory cytokines in peripheral blood mononuclear cells(PBMC)from the healthy volunteers.【Methods】PBMC isolated from the healthy volunteers were cultured and stimulated by lipopolysaccharide(LPS,the final concentration being 1μg/mL)or LPS+phytohemagglutinin(PHA,the final concentration being 25?μg/mL),and were also treated with or without TA(the final concentration being 5.4?ng/mL)simultaneously.The contents of tumor necrosis factor α(TNF-α),interleukin 4(IL-4)and interleukin 10(IL-10)in the supernatant of the culture were measured by enzyme-linked immunosorbent assay(ELISA).【Results】TA inhibited the production of TNF-α and IL-10 in PBMC culture stimulated by LPS or LPS+PHA,and the inhibitory effect of TA on IL-10 was stronger than on TNF-α,but had no effect on the production of IL-4.【Conclusion】TA has an inhibitory effect on the inflammation-promoting cytokine of TNF-α and anti-inflammatory cytokine of IL-10,which keeps the balance of inflammation-promoting and anti-inflammatory actions.
