Cultivation and identification of endothelial cells in vitro from human umbilical vein

Abstract

Objective To explore the primary culture method and summery the identified ways of human vascular endothelial cells(ECs) in vitro from umbilical vein.Methods ECs in umbilical vein were digested separated and subcultured with trypsin.And morphologic observation and evaluation of ECs were made with light microscopy,electron microscopy and immunohistochemistry.Results Enough ECs were obtained from digestive liquids,the monolayer cells were formed after7-10 days.The ECs arrayed like pitching stone under light microscopy;Ⅷ factor in the ECs showed positive reaction by immunohistochemistry.Weibel-Paladecorpuscles was in the cell plasm under electron-microscopy which were identified as ECs.Conclusion The digestion of pancreatin perfusion is an effective method to gain ECs,and this method provides experimental model for vascular ECs research.
