Culture,observation and identification of endothelial cells from human umbilical vein in vitro

Abstract

Objective To establish the model of human vascular endothelial cells (ECs) to provide important experimental method for pathogenesis research of atherosclerosis in vitra.Methods ECs in human umbilical vein (HUVECs) were digested by 0.125% trypsin+0.01% EDTA or 0.1%Ⅱtype collagenase.Morphologic observation and identification of HUVECs were made with light microscopy, electron micro-scopy and immunohistochemistry.Results The primary cultured HUVECs started to grow attaching to flask about 1 hours after inoculated.The HUVECs arrayed like pitching stone under light microscopy;Ⅷfactor in HUVECs showed positive reaction by immunohistochemistry;W-P body was in the cells plasm under electron-microscopy.All these cells were identified as ECs.Conchlsion High purity ECs are obtained with enzyme perfusion.This method could provide experimental model for vascular EC research.
