Effects of Interleukin-21 on Apoptosis of Jurkat Cells and Expression of WAVE-1

Abstract

Objective To study the effects of Interleukin-21(IL-21) on apoptosis of Jurkat cells and the probable functions of WAVE-1 in acute leukaemia(AL).Methods MTT was used to investigate the proliferation of Jurkat cells,flow cytometry(FCM) was used to investigate the effects of IL-21 on apoptosis of Jurkat cells.The expression of WAVE-1 in Jurkat cells treated with different concentraction of IL-21 was examined by RT-PCR.Results The growth of Jurkat cells were inhibited by IL-21 in the dose dependent manner(P0.05).FCM showed that the apoptosis rates of cells in experimental group were higher than those in control group,with significant difference(P0.01 or 0.05).The expression rates of WAVE-1 were decreased by the treatment with the increasing concentrations of IL-21(P0.05).Conclusion IL-21 can restrain the human leukemic cell growth and induce cell apoptosis,which may be related to the expressions of WAVE-1 decreased.
