Abstract
We aim to investigate the effects of bisindolylmaleimide(Bis) Ⅷ on the activation,proliferation,cell-cycle and apoptosis of Jurkat T cells,as well as to elucidate the mechanism of the process.Jurkat cells were divided into blank control group and Bis Ⅷ-treated groups.The expression level of CD69 on the surface of Jurkat T cells was evaluated by the technique of fluorescinconjugated monoclonal antibody double labeling;MTT method was used to analyze the proliferation of Jurkat T cells;the growing situation of Jurkat T cells was observed using microscope;PI staining and flow cytometry were used to analyzed the cell cycle;the apoptosis of Jurkat T cells was measured by Calcein staining.The experiment result showed that the expressions of CD69 of Jurkat T cells treated with different concentrations (5 μmol/L,10 μmol/L,20 μmol/L) of Bis Ⅷ decreased (P 0.05),and Bis Ⅷ could significantly inhibited the proliferation of Jurkat T cells in a dose-dependent manner (P 0.01).With the growing of the concentration of Bis Ⅷ,the cell cycle of Jurkat T cells was arrested in G0 /G1 phase.Furthermore,the Bis Ⅷ promoted the apoptosis of the Jurkat T cell in a dose-dependent manner.Thus,Bis Ⅷ can effectively inhibit the biologic behavior of Jurkat T cells,so we can conclude that Bis Ⅷ has potential to be a medicine of acute leukemia.
