Abstract
Objective:To investigate the effects of ganlike on the mRNA expression of MMP-2 and TIMP-2 in the rats with experimental hepatic fibrosis,and to explore the possible mechanism of ganlike's anti-hepatic-fibrosis.Methods:The hepatic fibrosis model was induced in the experimental rats by subscutaneous injection of 40% CCl 4 After hepatic fibrosis model being built,the rats in the ganlike groups were respectively given ganlike at the different dosages,the rats in the normal control and the hepatic fibrosis model group were given the same volume of saline.At the end of the eighteenth week,all the rats were killed.The severity of inflammation and fibrosis in all rats'liver tissues were measured by semiquantitative scoring.At last,The mRNA expression of MMP-2 and TIMP-2 in liver tissues were detected by in situ hybridization.Results:(1)Compared with the normal control group,the severity of inflammation scoring and hepatic fibrosis scoring in the liver tissues of hepatic fibrosis model group showed significant difference(P0 01);Compared with the hepatic fibrosis model group,the severity of inflammation scoring and the liver fibrosis scoring in the liver tissues of all the ganlike groups became obviously lighter (2)Compared with the normal control group,the expression of MMP-2 and TIMP-2 mRNA in liver tissues of the hepatic fibrosis model group were obviously higher,which showed significant difference;after the treatment,the expression of TIMP-2 in the different dosages of ganlike were obviously lower,the expression of MMP-2 showed no significant influence(P0 05).Conclusion ①Ganlkie may reduce the severity of inflammation scoring and hepatic fibrosis.②The mRNA expression of MMP-2、TIMP-2 in the liver tissues of the experimental hepatic fibrosis was enhanced,so MMP-2、TIMP-2 may be improtant factors in treatment of hepatic fibrosis.③Ganlike may downregulate the mRNA expression of TIMP-2 in the liver tissues of the experimental hepatic fibrosis,which may be the possible mechanism of ganlike's anti-hepaticfibrosis.
