Abstract
Taking hypocotyls of Medicago variaGannong No.2 as the explants and MS as basic medium,the effects of different addition concentration of 2,4-D,NAA,KT,BA,sucrose and the inducing time of callus on the callus induction,somatic embryogenesis and shoot differentiation were studied.The result showed that 3 mg/L 2,4-D+1.5 mg/L NAA was the optimum hormone combination,the proper action time of 2,4-D was 15 days,and the optimum culture medium for shoot differentiation was MS+0.5 mg/L KT+0.1 mg/L NAA supplemented with 7 g/L agar and 2 g/L sucrose.
