Determination of Palmatinechloride and Berberine Hydrochloride In Coptis chinensis Franch by HPLC and Fluorescence Detection

Abstract

OBJECTIVE To establish an HPLC method for determining the contents of palmatinechloride and berberine hydrochloride in Coptis chinensis Franch. METHODS The condition of detection were: Agilent C 18( 250mm ×4. 6mm,5μm),the mobile phase consisted of acetonitrile and 15mmol·L-1sodiumdodecylsulfate phosphate buffered saline( 0. 05mol·L-1Potassium Phosphate Monobasic,adjust with phosphoric Acid to a pH of 4. 0)( 40∶60),the FLD excitation wavelength was 354nm,the emission wavelength was 533nm. RESULTS The calibration curves were linear in the range of 1. 798 ~ 179. 8μg·mL-1for palmatinechloride( r = 1. 000); The calibration curves were linear in the range of 1. 762 ~ 176. 2μg·mL-1for berberine hydrochloride( r = 0. 9995). The average recoveries of palmatinechloride were 99. 98%( RSD = 3. 8%); the average recoveries of berberine hydrochloride were 99. 12%( RSD = 1. 4%). CONCLUSION The method is simple,accurate,reliable and can be used for the quality control of palmatinechloride and berberine hydrochloride in Coptis chinensis Franch.
