【Objective】 The suitable technology of ethanol extraction in the flavone from leaves in Neosinocalamus affinins was studied.The differences of flavone content in Neosinocalamus affinins from different regions of Sichuan were revealed,and the theoretical evidence was provided for culturing Neosinocalamus affinins with the high flavone content.【Method】 The suitable techniques of flavone extraction were studied through four factors,including ethanol concentration,ratio of sample with ethanol solution,extraction temperature and time,using the mature leaves of Neosinocalamus affinins in Southwest University of Science and Technology.The design of orthogonal experiment L9(34) was used to select the suitable techniques of extracting flavone from the leaves of Neosinocalamus affinins.The flavone content was determined by NaNO2-Al(NO3)3 colorimetric method.The flavone of the leaves,young shoots,old shoots in Neosinocalamus affinins in different regions of Sichuan was extracted through the suitable techniques.【Result】(1)The effect of ethanol concentration on the flavone extraction of leaves from Neosinocalamus affinins was significant and higher than that of the others.The second was the time extracting flavone of leaves.There was no significant ratio of samples with ethanol solution on the flavone extraction.(2)The flavone content of mature leaves in Neosinocalamus affinins was higher than that of young and old shoots through the comparative analysis of flavone contents from the mature leaves and young shoot and old shoots.(3)The differences of flavone content among them were found in different regions.The flavone contents of mature leaves from MSQ and YAL were the highest and the lowest respectively.The flavone contents of young shoots from DZD and old shoots from YBC were the highest.【Conclusion】 The techniques of flavone extraction were suitable under the ratio of 20 folds of 70% of ethanol with 1 fold of sample at 70 ℃ for 2 hours.The leaves of Neosinocalamus affinins in MSQ were suitable for extracting the flavone.
