Abstract
Objective To investigate the correlation between single nucleotide polymorphisms(SNP) of-1154G/A in VEGF gene and the onset risk of lung cancer of Han people population in north China.Methods The case-control study based on hospital was used,and the venous anticoagulated blood specimens of 200 patients with lung cancer and 204 healthy subjects were collected,meanwhile,the patients' medical history and the data about the healthy subjects were recorded.The genomic DNA was extracted by using proteinase K digestion followed by a salting out procedure.Polymorphisms of VEGF gene were analyzed by PCR-restriction fragment length polymorphism analysis(PCR-RFLP) and primer-introduced restriction analysis PCR(PIRA-PCR).Results There was no significant difference in the genotype of 1154G/A in VEGF and allele gene frequency distribution between lung cancer group and control group(P0.05).As compared with G/G genotype,the genotype with A allele gene(G/A+A/A) could reduce the onset risk of lung cancer in non-smokers.No correlation was found between SNP of 1154G/A in VEGF and onset risk of lung squamous-celled carcinoma,pulmonary adenocarcinoma,small cell lung cancer.Conclusion VEGF-1154G/A SNP may be correlated with the onset risk of lung cancer in non-smokers,and the genotype with A allele gene(G/A+A/A) can reduce the onset risk of lung cancer in non-smokers,as compared with G/G genotype.
