Abstract

[Objective] The aim was to find the appropriate concentration of kanamycin to screen the transformant of Portulaca oleracea L.[Method]The effects of kanamycin on the induction and growth of callus were researched by adding the different concentrations of kanamycin in Callus induction media and proliferation of callus culture.[Result]Kanamycin had a great effect on the callus induction,and it was severely inhibited by the concentration of 20 mg/L kanamycin.Its callus still grew at the concentration of 100 mg/L,but grew slowly and the degree of browning also increased with the increasing concentration of kanamycin.[Conclusion]20 mg/L kanamycin could be used as the selection pressure in the callus induction phase for the genetic transformation of Portulaca oleracea L.If callus was used as transformational receptor,80 mg/L kanamycin could be used as the selection pressure in the initial screening.
