Abstract
A rapid and efficient method for the quantitative determination of cholesterol in different kinds of eggs by high performance liquid chromatography(HPLC) with evaporative light-scatter detector(ELSD) was established.C18 chromatographic column served as the stationary phase.The mobile phase was methanol flowing at the rate of 1 mL·min-1 with the column temperature 25℃.The nitrogen gas flow rate of the ELSD was 2 L·min-1 with the temperature of the drift tube 85℃.A satisfactory linearity was obtained from 4.008~40.080 μg of cholesterol with a good correlation coefficient(R=0.9991).The recoveries for the low,middle and high concentration of the samples were 95.4%~99.1%.This method was found to be conveniently operated,rapid,efficient and accurate,as well as precise and reliable.
