Abstract

Objective:To investigate the anti-tumor effect of ampelopsin(APS) in vitro.Methods:The growth inhibition was analyzed by MTT and colony forming experiment in the lung cancer A549.The effect of APS on the expression of p53 mRNA in the human lung cancer cell line A549 was analyzed by RT-PCR.Results:Ampelopsin could obviously inhibit the growth of A549 cells cultured in vitro,with IC50 = 9.8 μg · mL -1 in MTT assay.The inhibition ratio in colony forming experiments were 100%,91.6%,64.5% and 23.4%,respectively,when dosages were 14.84,9.90,6.60 and 4.40 μg · mL-1.APS can significantly improve the expres-sion of p53 gene at 25 μg · mL -1.Conclusion:Ampelopsin can inhibit the growth of A549 in vitro remarkably,and the effect on expression of p53 gene is one of its anti-tumor mechanisms.
