Abstract

OBJECTIVE 12 dogs were divided into two groups for the study of the bioavailability of componund theophylline-ketofen sustained-release tablet(CTKSRT) vs theophylline controled-release tablet(TCRT) as reference drug.METHODS Both drugs were administered 200 mg orally. The blood plasma concentration of theophylline were assayed by HPLC columm switching method. The pharmacokinetic parameters of CTKRST and TCRT were calculated by 3p87 program.RESULTS There were no significant difference between the pharmacokinetics parameter of CTKRST and TCRT.AUC:236±34.80 and 207.88±31.80 mg·L -1·h -1;C max: 12.99±3.11 and 11.55±2.83 mg·L -1;T max:6.88±0.98 and 7.00±0.89 h,respectively. The relative bioavailability of CTKSRT was 113.76%.CONCLUSION The two formulations both are extended release and with bioequivalent in dogs administered orally.
