Abstract

Objective To study the effect of Huomai tongluo decoction(HTD) on Caspase-3 expression in brain tissue of the cerebral ischemia and reperfusion rats to explore the mechanism of HTD effect on damaged brain.Methods Wistar's rats were randomly divided into normal control group,ischemia-reperfusion group,low-dose of HTD group,middle-dose of HTD group,high-dose of HTD group and nimodipine positive control group.Using a modified suture method,the cerebral ischemia and reperfusion model of middle cerebral artery occlusion(MCAO)was established.HE staining was used to observe morphology changes in rat brain tissue,immunohistochemistry and Western blot to assay Caspase-3 expression in brain tissue qualitatively and quantitatively.Results Caspase-3 expression level was significantly increased in ischemia-reperfusion group than in normal control group( 0.01),but significantly reduced in the HTD groups compared with ischemia-reperfusion group group(0.01),however,with no significant difference HTD group compared with nimodipine group.Conclusion The protective effect of HTD might be related to the inhibition of apoptosis in the brain of ischemia-reperfusion rat.
