Abstract
Objective:It is to observe the influence of Danxing Tongluo decoction(DXTLD)on nerve cell Caspase-3 expression in rats with cerebral ischemia reperfusion injury and to study its nerve protection mechanism.Methods :Forty healthy male Wistar rats were randomly divided into 5 groups:sham operation group,ischemia/reperfusion(I/R)group,DXTLD high dosage group,DXTLD low dosage group and Nimodipine(NIMO)group,8 rats per group.DXTLD groups were given to DXTLD at dose of 15mL/kg,7.5mL/kg;NIMO at 10mg/kg,one time a day.Saline were given to sham operation group and I/R group at 10mL/kg weight.After 7-days of intragastried.The rats of all the groups except sham group were made into cerebral ischemia reperfusion injury models by thread embolism method.All the rats were killed and their brains were taken out after 2h ischemia and 24h reperfusion injury.The Caspase-3 expression in brain tissue was measured with immunohistochemical method and was compared with the change of pathohistology.Results:DXTLD could obviously decrease the Caspase-3 expression in cerebral cortex and hippocampus and relieve nerve cell injury.Conclusion:DXTLD can protect brain tissue from cerebral ischemia reperfusion injury and improve the nerve function.The mechanism may be related with the decreasing of Caspase-3 expression.
