Abstract
Objective To observe the Huomaitongluo decoction on nuclear factor- κB(NF- κB) and interleukin-1β(IL-1β) proteins expression in cerebral ischemia and reperfusion rat brain, and to explore the protective mechanism of the Huomaitongluo decoction. Methods The rat middle cerebral artery occlusion(MCAO) model of cerebral ischemia and reperfusion was established by modified suture method. The rats were devided into normal control group,ischemia-reperfusion group,Huomaitongluo decoction low-dose group and the middle dose group and high-dose group, and Ginkgobiloba positive control group respectively.Huomaitongluo decoction and Ginkgobiloba were administered orally 24h after reperfusion, two times a day for three consecutive days.Western blot was used to detect the expression of NF- κB and IL-1β proteins. Results The expression levels of NF-κB and IL-1β proteins in brain tissue of rat were increased in ischemia-reperfusion group than in control group(P0.01),but lower in Huomai tongluo decoction low-dose group and middle dose group and ginkgobiloba group than in ischemia-reperfusion group(P0.01 or P0.05),but with no significant difference between the high dose group and ischemia-reperfusion group. Conclusion The treatment effect of Huomaitongluo decoction on the ischemia-reperfusion damage might be related to the downexpression of NF- κB and IL-1βin brain tissue.
