Abstract
Objective To investigate the mechanism of Jurkat cell(human myelocytic leukaemia) apoptosis and cell cycle alteration induced by Sodium pheny-acetate(NaPA).Method MTT and FACS were applied to detect Jurkat cell proliferation inhibition rate and cell cycle alteration treated by NaPA.Electron microscope was used to observe subcellular structure alteration of Jurkat cell.Results NaPA could inhibit Jurkat cell proliferation rate which varied from 27%~58% with dose dependent.FACS analysis showed cell in stage G0/G1 increased and in stage S decreased with the relative increase of stage G2/M,Jurkat cell apoptosis rate increased.Mitochondria swelling and karyopycrosis could be observed under the electron microscope.Conclusion NaPA can inhibit Jurkat cell cycle progression from stage G1 to S and induced Jurkat cell apoptosis.
