Effect and Mechanism of Tumor Necrosis Factor on Hepatic Injury Rats

Abstract

Objective: To study the effect and mechanism of tumor necrosis factor inducing hepatic injury. Methods: 24 male Sprague Dawley rats were randomly devided into normal saline control group (Ⅰ)and TNF group(Ⅱ). The rats in TNF group were inject with TNF of 1.6×10 6 u/kg through vein. ALT?AST?SOD, and MAD in serum at 0, 2, 4, and 8 hours after injection were determined. The liver tissues were observed with light and electron microscope at 8 hours. Results: (1) ALT and AST in TNF group increased significantly compared with those before injection ( P 0.05).Liver tissues were found damage, including hepatocellular degeneration and necrosis. (2) MDA of TNF group increased significantly but SOD of the group decreased significantly (both P 0.05). Conclusion:(1)TNF induces obviously hepatic injury;(2)Free radicals play an important role in TNF mediated cytotoxicity.
