The effect of tumor necrosis factor- α monoclonal antibody in liver of rats on acute hemorrhagic shock

Abstract

Objective To probe the role of tumor necrosis factor-α(TNF-α) in liver injury of rats on acute hemorrhagic shock and the protective effect of tumor necrosis factor-α monoclonal antibody(TNF-α McAb).Methods Twenty-four male Sprague-Dawley rats were randomly divided into three groups: control group(A),shock+lactated Ringer's solution resuscitation group(B) and shock+TNF-αMcAb resuscitation group(C).Through the femoral artery bleeding,animal models of acute hemorrhagic shock were established.B group with lactated Ringer's solution resuscitation,C group with lactated Ringer's solution resuscitation containing TNF-αMcAb(3mg/kg),while the A group under the same conditions without loss of blood.The levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST) and TNF-α were detected in each group.The contents of malondialdehyde(MDA) and superoxide dismutase(SOD) in liver tissue were measured.The pathological changes of liver tissue were observed under light and electron microscopy.Results The serum levels of ALT,AST,TNF-α and the content of MDA in liver tissue increased in B and C group compared with A group,while the content of SOD decreased.The serum levels of ALT(343.63 ± 35.61)U/L,AST(748.75 ± 49.76)U/L,TNF-α(99.38 ± 13.16)pg/mL and the content of MDA(26.33±1.30)nmol/mgProt in liver tissue in C group were much lower than those in B group,but the content of SOD(510.14±47.44) U/mg Prot was increased.The changes of liver tissue histopathology were minor in C group.Conclusion TNF-α may be one of an important factor in liver injury induced by acute hemorrhagic shock.Use of TNF-αMcAb resuscitation can reduce liver injury and has a protective effect in rats with acute hemorrhagic shock.
