Abstract
A HPLC metheod was developed for the determination of uracil and uridine in the manufacturing process of 5' - deoxy - 5 - fluoro - uridine. Reversed - phase chromatographic separation was carried out on a Spherisorb C18 column (150 mm × 4.6 mm) .A mixture constituted by acetonitril - pH4.5,0.02 mol/L phosphate buffer (2:98) was used as the mobile phase. Complete separation was achieved between uracil and uridine under optimum conditions. No interference from other components in samples was found. The recoveries were as high as 99.10% - 100.4% for uracil and uridine. The proposed method is simple, accurate and reproducible.
