Abstract
Objective Objective To investigate the diagnostic efficacy of purified 14-33 ku antigens in immunodiagnosis of clonorchiasis. Methods The 14-33 ku antigens was purified from the soluble adult worm antigens of clonorchis sinensis by 12% SDS-PAGE and elution, and was used to detect specific IgG antibodies in sera of patients with clonorchiasis, paragonimiasis, schistosomiasis japonica, fasciolopsiasis and healthy persons by ELISA.The soluble adult worm antigen(AWA) and the soluble degrease adult worm antigen(DAWA) were used in the control test. Results The sensitivities were 76.2% by 14-33 ku antigens and 100% by AWA and DAWA for detecting 64 cases of clonorchiasis. The cross reaction rates were 8.0%,3.5%,0 respectively by 14-33 ku antigens for detecting 25 cases of paragonimiasis,85 cases of schistosomiasis japonica and 27 cases of fasciolopsiasis, whereas the cross reaction rates were 80.0%,62.4%,14.8% by AWA and 64.0%,55.3%,7.4% by DAWA respectively. The false positive rates were 0,5.5% and 3.1% respectively by 14-33 ku antigens,AWA,DAWA for detecting 127 cases of healthy persons. Conclusion Conclusion The purified 14-33 ku antigens of clonorchis sinensis has higher specificity and lower sensitivity in detecting the specific IgG of clonorchiasis compared to the soluble raw antigens of clonorchis sinensis.
